ThepropertiesofexosomessecretedbyhumanumbilicalcordblooddendriticcellsanditsenhancementofCTLcytotoxicityDUYing1,CHENShao-qian2,HUANGYu-min1,DONGZi-ming3*1DepartmentofMicrobiologyandImmunology,CollegeofBasicMedicalSciences,2DepartmentofHematology,FirstAffiliatedHospital,3DepartmentofPathophysiology,CollegeofBasicMedicalSciences,ZhengzhouUniversity,Zhengzhou450052,ChinaAIM:Toinvestigatethepropertiesofexosomessecretedbydendriticcells(DCs)derivedfromhumanumbilicalcordbloodmononuclearcells(HUBMCs)anditsenhancementofCTLcytotoxicity.METHODS:TheHUBMCswereseparatedfromhumanumbilicalcordblood(HUCB).ThegenerationofDCswereinducedfor12daysinmediumcontainingSCF,IL-4andGM-CSF.Tumorantigenextractedbyrepeatedfreeze-thawingwereaddedtoculturemediumandculturedsuccessivelyfor2days.TheDCswerecollectedandexosomeswereseparatedfromculturesupernatantthroughcentrifugationat100000g.Theexosomes-associatedpropertieswereanalyzedbyFCM,SDS-PAGE,transmissionelectronmicroscope(TEM)andMTTcolorimetry.RESULTS:Afterbeinginducedfor12dayswithSCF,IL-4andGM-CSFthenumberofDCshada10-foldincreaseanddisplayedtypicallymorphologicalcharacteristicsofDCs.FCManalysisshowedthattheDCsloadedwithtumorantigenexpressedhighlyMHC-I,MHC-II,CD40,CD80,CD86,CD11candCD54.ExosomescouldbeseparatedfromculturesupernatantofDCsbyultracentrifugation.Mixedlymphocytereation(MLR)andcytotoxicityassayshowedthatexosomessecretedbyDCscouldeffectivelystimulateTcellproliferationandenhancetheircytotoxicity.CONCLUSION:AlargenumberofDCsareobtainedfromUBMCsandtheexosomesaresecretedfromtheminvitro.TheexosomescanpresenttumorantigentoTcellsandenhancethecytotoxicityofCTLs.[【Fund】：河南省重大科技攻关项目基金资助(No.001170209)
